    開發(fā)TRAP 標(biāo)記的新策略

    A Novel Strategy for TRAP Marker Development

    • 摘要: 煙草作為重要的經(jīng)濟(jì)作物和生物工程研究領(lǐng)域的模式植物,,由于基因組龐大,、結(jié)構(gòu)復(fù)雜,、重復(fù)序列較多以及缺少理想的分子標(biāo)記,致使大量重要農(nóng)藝,、品質(zhì)性狀缺少分子水平上的遺傳研究。因此開發(fā)適合煙草的分子標(biāo)記對其遺傳研究具有重要意義,。本研究在以往TRAP 標(biāo)記開發(fā)經(jīng)驗的基礎(chǔ)上,,探索了一種直接利用EST-SSR 引物作為固定引物開發(fā)TRAP 標(biāo)記的新方法,,降低了TRAP 標(biāo)記的開發(fā)難度和成本,,在短期內(nèi)能開發(fā)出豐富的TRAP 標(biāo)記組合,。本研究開發(fā)了首套煙草TRAP標(biāo)記,結(jié)果顯示其擴(kuò)增帶型理想,,多態(tài)性較高,,并在普通煙草品種中篩選到一條類香料煙特殊高香氣基因型特征帶,顯示出良好的應(yīng)用前景,。

       

      Abstract: Tobacco is an important agricultural model plant that plays a significant role in the economies of many countries. Furthermore, tobacco is considered to be one of important model systems in plant biotechnology. However, the genetic studies, such as molecular marker development, linkage map construction and gene cloning of tobacco, are relatively backward. The reasons are most likely that tobacco is a tetraploid species with the biggest genome size in Solanaceae and complex genome structure. Therefore, it is important to develop excellent molecular markers for tobacco genetic study. Then, a new strategy for tobacco-specific TRAP molecular marker development was adopted in this study, in which the primers of tobacco EST-SSR was shared directly for the fixation primers of TRAP markers, and the first set of tobacco TRAP markers were developed. Compared with previous studies, the new strategy has several advantages, such as shorter process, lower cost and more polymorphism than EST-SSR. Moreover, the further studies confirm that the new strategy is useful for tobacco molecular genetic studies because of the good PCR amplification and a high level genetic diversity. In addition, a co-dominant heredity TRAP characteristic band of special aroma of oriental tobacco-like genotype was found, which can be transformed to SCAR marker or used in marker assisted selection (MAS) directly.
