    Fermentation Conditions of Antagonistic strain Tpb55 of Bacillus subtilis against Tobacco brown spot

    • 摘要: 通過單因素和正交實驗相結合的方法,,研究了枯草芽孢桿菌(Bacillus subtilis)Tpb55液體發(fā)酵培養(yǎng)液和發(fā)酵條件,,篩選出最佳培養(yǎng)液配方為:葡萄糖1%(10 g)、蛋白胨0.3%(3 g),、MgSO4 0.05%(0.5 g),、酵母膏0.15%(1.5 g)與牛肉膏0.15%(1.5 g)的混合物,最適發(fā)酵條件為:溫度31℃,,初始pH7.0,,發(fā)酵時間72 h。

       

      Abstract: Bacillus subtilis Tpb55 was isolated from tobacco phyllosphere and had antagonistic activity against to tobacco brown spot. Cultivation medium of Tpb55 was researched by single factor and orthogonal experiments. The fermentation conditions tested included fermentation medium, pH value before sterilized, fermentation temperature and fermentation time. The results showed that the optimal cultural liquid consisted of 1% glucose (10 g), 0.3% peptone (3 g), 0.05% Mgso4 (0.5 g), 0.15% yeast extract (1.5 g) and 0.15% (1.5 g) beef extract. The optimal fermentation conditions of stain Tpb55 were predicted as below: temperature 31℃, pH7.0 and 72h, agitation 180r/ min.
